Is it possible to work with my purchase code in localhost , without de-register from online site ?

This depends from case to case. Some items requires one activation only without deactivation possibility.
Others lets you do this without problems.
But there are few other items where you need to contact the theme author to de-register the key used on localhost in order to be able to re-register it on your live site.
